實(shí)驗(yàn)步驟
1．首先將內(nèi)層滅菌桶取出，再向外層鍋內(nèi)加入適量的水，使水面與三角擱架相平為宜。
2．放回滅菌桶，并裝入待滅菌物品。注意不要裝得太擠，以免防礙蒸汽流通而影響滅菌效果。三角燒瓶與試管口端均不要與桶壁接觸，以免冷凝水淋濕包口的紙而透入棉塞。
3．加蓋，并將蓋上的排氣軟管插入內(nèi)層滅菌桶的排氣槽內(nèi)。再以兩兩對(duì)稱的方式同時(shí)旋緊相對(duì)的兩個(gè)螺栓，使螺栓松緊*，勿使漏氣。
4．用電爐或煤氣加熱，并同時(shí)打開(kāi)排氣閥，使水沸騰以排除鍋內(nèi)的冷空氣。待冷空氣*排盡后，關(guān)上排氣閥，讓鍋內(nèi)的溫度隨蒸汽壓力增加而逐漸上升。當(dāng)鍋內(nèi)壓力升到所需壓力時(shí)，控制熱源，維持壓力至所需時(shí)間。本實(shí)驗(yàn)用 1．05kg/cm2，121．3℃，20分鐘滅菌。
5．滅菌所需時(shí)間到后，切斷電源或關(guān)閉煤氣，讓滅菌鍋內(nèi)溫度自然下降，當(dāng)壓力表的壓力降至0時(shí)，打開(kāi)排氣閥，旋松螺栓，打開(kāi)蓋子，取出滅菌物品。如果壓力未降到0時(shí)，打開(kāi)排氣閥，就會(huì)因鍋內(nèi)壓力突然下降，使容器內(nèi)的培養(yǎng)基由于內(nèi)外壓力不平衡而沖出燒瓶口或試管口，造成棉塞沾染培養(yǎng)基而發(fā)生污染。
6．將取出的滅菌培養(yǎng)基放入 37℃溫箱培養(yǎng)24小時(shí)，經(jīng)檢查若無(wú)雜菌生長(zhǎng)，即可待

Experimental steps

After the cold air is compley drained off, close the exhaust valve so that the temperature inside the pan gradually increases with the increase of steam pressure. When the pressure in the pan rises to the desired pressure, the heat source is controlled and the pressure is maintained until the desired time. The experiment with 1.05kg / cm2, 121.3 ℃, 20 minutes sterilization.
5. Turn off the power or turn off the gas to allow the temperature in the autoclave to drop naturally. When the gauge pressure drops to 0, open the vent valve, loosen the screw, open the lid and remove the sterilized items. If the pressure has not dropped to 0, open the exhaust valve, the pressure will be due to a sudden drop in the pot, so that the media inside the container due to internal and external pressure imbalance and out of the flask mouth or test tube mouth, resulting in tampon contaminated medium occurred Pollution.
6. Will be taken out of sterile medium incubated in 37 ℃ incubator 24 hours, after checking if no bacteria growth, you can wait
