方法如下:
1.人IgG聚合物的制備 在5ml含40mg/ml人IgG的0.1mol/L pH7.0磷酸緩沖溶液中,加入2.5%戊二醛溶液1ml,邊加邊攪,5min即出現(xiàn)混濁,逐現(xiàn)大塊膠塊,放置30min后,用研缽將凝膠磨細(xì),繼用1.0mol/L pH7.0磷酸緩沖溶液反復(fù)洗滌3次,末次加蒸餾水至20ml,即為人IgG聚合物懸液。
2.免疫吸收法
將待吸收的抗SIgG血清加入待量IgG聚合物懸液,置室溫?cái)嚢?0min,離心沉淀,上清液即稀釋1倍的純化抗SIgA血清。如用IgG聚合物作少量分次吸收,其效果更好。
(八)伊文氏藍(lán)(Evans blue)襯染法
用0.01%伊文氏藍(lán)的0.01mol/L pH7.2PBS稀釋熒光抗體,可將背景細(xì)胞和組織染色,呈紅色熒光,與特異性黃綠色熒光形成鮮明的對(duì)比,減少了非特異性熒光,宜作常規(guī)應(yīng)用。伊文氏藍(lán)一般先配成1%溶液,保存于4℃,用前再稀釋至0.01%用以和然釋熒光抗體。 此外,還可以用胰酶消化組織切片或用10%牛血清蛋白封閉法等消除非特異性染色,提高特異性染色。
